Why D-neuron? Importance in schizophrenia research
نویسنده
چکیده
Recent pharmacological discovery on trace amineassociated receptor, type 1(TAAR1) has emphasized importance of trace amines in pathogenesis of psychoses, such as schizophrenia. TAAR1 has many ligands, including tyramine, β-phenylethylamine (PEA), amphetamines, and 3’-iodothyronamine. Socalled D-neurons are putative producer of trace amines, endogenous ligands of TAAR1. The D-neuron is defined “the aromatic L-amino acid decarboxylase (AADC)-containing neuron, but not dopaminergic nor serotonergic”, i.e. not containing tyrosine hydroxylase nor tryptophan hydroxylase. AADC is an enzyme, also called dopa decarboxylase (DDC). The localization of D-neurons in the central nervous system has been specified into 15 groups, from the spinal cord (D1) to striatum (D15). We showed the decrease of D-neurons in D15 in postmortem brains of schizophrenia, where midbrain dopamine (DA) neurons are heavily innervated. Decrease of D-neurons may cause reduction of trace amines in the striatum, and may also decrease stimulation of TAAR1 on striatal terminals of ventral tegmental area (VTA) DA neurons. This might increase firing frequency of VTA DA neurons, and causes DA hyperactivity in the striatum and nucleus accumbens. In the present article, the author introduces the novel theory, “D-cell hypothesis”, for mesolimbic DA hyperactivity of schizophrenia. Some clinical and/or experimental evidences that support this hypothesis are mentioned. The D-neuron, as a trace amine producer, is a clue for elucidating pathogenesis of psychoses, as well as human mental functions. Thus, signal transduction of D-neurons should be investigated.

Perspective of D-Neuron (Trace Amine Neuron) Research: From Novel Therapeutic Strategies
Recent pharmacological studies have shown the importance of trace amine-associated receptor, type 1 (TAAR1), a subtype of trace amine receptors, to be a prospective target receptor for novel neuroleptics. Socalled D-neuron (trace amine (TA) neuron) is the ligand neuron of TAAR1. In 1984, Jaeger et al. specified D-neuron groups from D1 (the spinal cord) to D14 (the bed nucleus of stria terminali...
متن کاملA Mechanistic Approach to Preventing Schizophrenia in At-Risk Individuals
In this issue of Neuron, Schobel et al. (2013) report that excess hippocampal glutamate may cause transition to psychosis in individuals at high risk for schizophrenia. Their data has implications for prevention strategies and why stress often precipitates this transition.
متن کاملThe importance of studying metadiscourse
This essay focuses on metadiscourse, a name for elements of texts that convey meanings other than those that are primarily referential. The essay provides some theoretical background to the study of metadiscourse, briefly reviews a taxonomy of metadiscourse, and explores four reasons why the study of metadiscourse is interesting and important: (a) Such study shows how ...
متن کاملDe Novo CNVs in Bipolar Disorder: Recurrent Themes or New Directions?
In this issue of Neuron, Malhotra and colleagues report an enrichment of de novo copy number variants in bipolar disorder and schizophrenia when compared with those of controls. The study highlights the importance of a genetic model involving rare and disruptive variants to further our understanding of complex neuropsychiatric traits.
متن کاملNMDA receptor hypofunction, parvalbumin-positive neurons, and cortical gamma oscillations in schizophrenia.
Gamma oscillations appear to be dependent on inhibitory neurotransmission from parvalbumin (PV)-containing gamma-amino butyric acid neurons. Thus, the abnormalities in PV neurons found in schizophrenia may underlie the deficits of gamma-band synchrony in the illness. Because gamma-band synchrony is thought to be crucial for cognition, cognitive deficits in schizophrenia may reflect PV neuron dy...
